The provision of key biological data generated to high quality standards within short cycle times to drive compound optimization, is a service available to all medicinal chemistry partners of Evotec. Various assay formats including binding and catalytic assays for receptor and enzyme targets respectively, as well as functional data from cellular models are available. Off-the-shelf selectivity assays are available in the target areas kinases, GPCRs, ion channels, protease and nuclear hormone receptors. Many of the biochemical chemistry support assays are conducted with radiometric read-outs like filter binding or SPA and LC/MS based read-outs for enzyme targets. In-depth studies for cellular efficacy are also available with more complex cellular models such as primary cells and also more sophisticated read-outs such as imaging or qPCR.
In order to rationalize inhibitor action, Evotec offers mode-of-inhibition studies which include reversibility, competitive versus uncompetitive, and non-competitive inhibition with respect to substrates, experimental Ki determination, slow on-rate inhibition and others.
